The Murciélago (2001–2010) replaced the Diablo and was the first new Lamborghini developed under Audi ownership. Designed by Luc Donckerwolke, it has scissor doors, a steel frame with mostly carbon-fibre body panels, a V12 mounted lengthwise behind the seats and four-wheel drive with a viscous centre coupling. The gearbox is a six-speed manual or the six-speed e-gear automated manual.
The first cars have a 6.2-litre V12 with 580 PS; the Roadster, with a soft top that is fitted by hand, followed in 2004. In 2006 the LP 640 brought a 6.5-litre engine with 640 PS and a facelift, and the LP 640 Roadster was shown at the end of that year. The last versions were the 670 PS LP 670-4 SuperVeloce (2009–2010), about 100 kg lighter, and the LP 650-4 Roadster, limited to 50 cars. In total 4,099 Murciélagos were built; the Aventador took over in 2011.

The original Murciélago (2001–2006) has a 6.2-litre V12 with 580 PS. The LP 640 (2006–2010) has a larger 6.5-litre engine with 640 PS and a facelift, with new front and rear ends, asymmetric side air intakes (the left one feeds an oil cooler) and a single exhaust outlet in the rear diffuser. Carbon-ceramic brakes became an option.
